产品介绍
氯离子荧光探针MQAE是美国AAT Bioquest生产的用于检测氯离子的荧光探针。氯化物指示剂,其荧光依赖于氯离子浓度。MQAE的荧光强度随着氯离子在细胞内的增加而成比例地减少。MQAE具有高水溶性和膜渗透能力,可以检测低于50 mM浓度的氯离子浓度而不受pH变化的影响。MQAE的检测灵敏度比SPQ更高,并且荧光效果也比SPQ要好一些,是一种目前最广泛应用的新型氯离子荧光探针。金畔生物是AAT Bioquest的中国代理商,为您提供最优质的氯离子荧光探针MQAE。

操作步骤
1. 用Krebs-HEPES缓冲液洗涤细胞3次。
2. 将5 mM的Cl探针溶液加入细胞液中,并在37℃下培养1小时。
3. 用Krebs-HEPES缓冲液洗涤细胞5次。
4. 使用带有图像分析系统的激光共聚焦显微镜检测细胞的荧光强度。
注1:MQAE的工作液可以配制于Krebs-HEPES缓冲液中;Krebs-HEPES缓冲液配方(20mM HEPES,128mM NaCl,2.5mM KCl,2.7mM CaCl2,1mM MgCl2,16mM glucose,pH7.4)
注2:MQAE标记细胞时的浓度通常为5-10mM。以约800-1000万cell/ml的细胞密度,在含有5-10mM MQAE的Krebs-HEPES缓冲液中孵育30-60分钟。通常一个探针的装载体系为100ul即可。
