简要概述

着色剂-ALL是美国AAT Bioquest生产的用于核酸和蛋白质差异染色的试剂。它可染色蓝紫色的RNA,蓝色的DNA和红色的蛋白质。在聚丙烯酰胺凝胶上可以检测到少至3ng(123 BP片段)的pBR322 / Hae III消化DNA和90ng tRNA。 PAGE凝胶在黑暗中染色,然后通过从染色溶液中除去凝胶并将其暴露于来自灯箱的光直至发生足够的脱色而脱色。通常通过将染料溶解在甲酰胺和缓冲液中来制备染色溶液。金畔生物是AAT Bioquest 的中国代理商,为您提供最优质的染色试剂。

产品说明书

操作方法

1.将10mg Stains-All加入10mL甲酰胺中,制成0.1%甲酰胺原液染色剂。

 

2.所有工作溶液如下:

1ml储备Stains-All

1ml甲酰胺

5ml异丙醇

100ml 3.0M Tris-HCl,pH8.8

12.9ml水

 

3.Stains

3.1.RNA  – 蓝紫色

3.2.DNA  – 蓝色

3.3.蛋白质 – 红色

3.4.各种酸性粘多糖

 

4.Gel电泳:

4.1将凝胶浸泡在有盖的容器中18至24小时,避光。

4.2将凝胶浸泡在水中,避光。

注1:曝光会产生暗黄色背景。

注2:脱色也可以通过将凝胶暴露于光下30分钟来完成。

产品说明书

用于染色细胞的样品方案

以下过程提供了一般准则,实际情况应根据您的特定应用进行修改。

 

1.制备5-10 mM DMSO储备溶液。应将未使用的DMSO储备溶液等分到单次使用的小瓶中并储存在-20℃,避光。

2.在生理缓冲液(如PBS,HBSS,HEPES)中使染料的工作浓度为5-20μM。最佳工作浓度必须根据经验确定。

3.将染料工作溶液(来自步骤2)的等体积(例如100μL细胞在生长培养基中)加入细胞中,并在室温或37℃下孵育细胞5至60分钟。

4.在将细胞暴露于实验诱导物之前,确定负载细胞样品的基线荧光强度。

5.阴性对照应评估如下:

5.1检查有和没有诱导物的染料和缓冲液/培养基的无细胞混合物的荧光。 在没有细胞外酯酶和其他氧化酶的情况下,荧光随时间的逐渐增加可能与自发水解,大气氧化和/或光诱导氧化有关。

5.2检查已经保持在生长培养基或缓冲液中的未处理(对照)负载细胞的荧光。在健康细胞中,氧自由基被细胞酶和/或天然抗氧化剂消除。 在染料加载恢复期后,健康细胞应表现出低水平的荧光,在实验期间相对稳定; 然而,可以观察到荧光逐渐增加(由于自动氧化)或减少(由于细胞中的染料损失或光漂白)。在没有任何刺激或诱导的情况下,健康的未经处理的细胞中的荧光突发可以指示细胞死亡或一些其他氧化事件的进展。

6.可以用叔丁基过氧化氢(TBHP)刺激阳性对照至终浓度~100μM(基于细胞的敏感性和反应增加或减少剂量)。

产品说明书

样品实验方案

简要概述

1.准备和添加标准品和样品(50μL)
2.准备并添加酪氨酸酶蓝工作溶液到标准品和样品孔(50μL)
3.将板在室温下孵育60至120分钟
4.检测510nm处的吸光度

 

溶液制备

1.储备溶液

所有未使用的储备溶液应分成一次性等分试样,并在制备后储存在-20°C。 避免反复冻融循环。
1.1酪氨酸酶原液(2000 U / mL):
将120μL测定缓冲液(组分B)加入酪氨酸酶标准品(组分A)中并充分混合。 注意:将未使用的酪氨酸酶原液储存在-20°C,一次性使用等分试样。

1.2酪氨酸酶蓝原液(100X):
将50μLDMSO(组分D)加入酪氨酸酶蓝(组分C)中并充分混合。 注意:将未使用的酪氨酸酶蓝原液在-20°C下以一次性等分试样储存。

 

2.标准溶液

酪氨酸酶标准
使用酪氨酸酶原液(2000 U / mL)和测定缓冲液产生400 U / mL终浓度的酪氨酸酶标准溶液(T1)。 然后进行1:2连续稀释以获得剩余的连续稀释的酪氨酸酶标准品(T2-T7)。 注意:井中标准品的最终浓度为2X。 注意:使用提供的标准,如果使用建议的浓度,可以重复生成2条标准曲线。

 

3.工作溶液

酪氨酸酶蓝工作溶液:
通过将5μL酪氨酸酶蓝原液(100X)加入1mL测定缓冲液(组分B)中进行1:100稀释并充分混合。

1.以下协议可用作指南,应根据需要进行优化。

2.根据测定缓冲液中的建议制备标准品和测试样品,分别加入50ul到微孔板中。

3.将50μL酪氨酸酶蓝工作溶液加入标准品和样品的孔中。

4.将反应在37℃孵育60至120分钟。 注意:反应可保持长达6小时。

5.用光吸收酶标仪在510 nm处检测吸光度。
