产品说明书
操作方案
标记寡核苷酸
1.准备以下试剂:
200mMTHPTA [tris(3-羟丙基三唑基甲基)胺]水溶液
100mM CuSO4水溶液
炔烃修饰的低聚水溶液(尽可能浓缩,例如>10 mg / mL)
100 mM抗坏血酸钠水溶液
10 mM染料叠氮化物水溶液或DMSO溶液(有关推荐的溶剂,请参阅我们的网站)
2.在反应前将CuSO4与THPTA以1:2的比例混合并充分涡旋几分钟。该溶液在冷冻时可稳定数周。
3.向炔烃改性的低聚物溶液中加入过量的染料叠氮化物(摩尔比为2-5当量)。
4.加入5当量的THPTA / CuSO4工作溶液(来自步骤1)。
5.加入10-30当量的抗坏血酸钠。
6.在室温下搅拌,涡旋或摇动反应混合物30-60分钟。
7.沉淀或纯化寡聚物。
标记肽
1.准备以下试剂:
200 mM THPTA配体水溶液
100mM CuSO4水溶液
炔烃修饰肽水溶液或DMF溶液(取决于您的肽溶解度,如果可能,> 10 mg / mL)
100 mM抗坏血酸钠水溶液
10 mM染料叠氮化物水溶液或DMSO溶液(有关推荐的溶剂,请参阅我们的网站)

标记炔烃有机小分子
1.准备以下试剂:

炔烃化合物水溶液或DMF溶液(取决于您的化合物溶解度,如果可能,> 10mg / mL,)
100 mM抗坏血酸钠水溶液
10 mM染料叠氮化物水溶液或DMSO溶液(有关推荐的溶剂,请参阅我们的网站)。
2.在反应前几分钟以1:2的比例孵育CuSO4与THPTA配体。当冷冻时,该溶液可稳定数周。

标记生物聚合物
1.准备以下试剂:
200 mM THPTA配体水溶液
100mM CuSO4水溶液
炔烃改性的水中生物聚合物(尽可能浓缩,例如> 5毫克/毫升)
100 mM抗坏血酸钠水溶液
10 mM染料叠氮化物水溶液或DMSO溶液(有关推荐的溶剂,请参阅我们的网站)。
2.在反应前几分钟以1:2的比例孵育CuSO4与THPTA配体。 该溶液在冷冻时可稳定数周。
3.向炔烃改性的生物聚合物溶液中加入过量的染料叠氮化物(负载比:5-20染料叠氮化物/炔烃)。
4.加入5摩尔当量(参考染料叠氮化物)的THPTA / CuSO4。
5.加入10当量的抗坏血酸钠(参见染料叠氮化物)。
6.在室温下搅拌,涡旋或摇动反应混合物30-60分钟。
7.通过凝胶过滤或透析纯化您想要的分子。
标记细胞,细胞裂解物或生物样品
1.准备以下试剂:
100mM THPTA配体水溶液
20mM CuSO4水溶液
300mM抗坏血酸钠水溶液
2.5mM炔烃或叠氮化物标记试剂水溶液或DMSO溶液
2.对于每个叠氮化物或炔烃修饰的细胞或细胞裂解物样品,将以下试剂加入1.5 mL微量离心管中,然后短暂涡旋混合。
50μL细胞或细胞裂解液样品
50μLPBS缓冲液
50μL5mM相应的染料叠氮化物水溶液或DMSO溶液(或染料炔烃)检测试剂
3.加入10μL100mM THPTA溶液,短暂涡旋混合。
4.加入10μL20mM CuSO4溶液,短暂涡旋振荡。
5.加入10μL300mM抗坏血酸钠溶液以引发点击反应,短暂涡旋混合。
6.保护点击反应不受光照,使其在室温下孵育30分钟。
7.点击标记细胞或细胞裂解物并准备用于下游处理和分析。
