简要概述

藻蓝蛋白(APC)是一种蓝藻藻螺旋藻中分离的藻胆蛋白。像其他藻胆蛋白一样,APC是荧光的,具有极高的吸收率和高量子效率。我们的APC-iFluor 800串联探针拥有一种独特的荧光团,具有用于光谱流式细胞术的IR发射。它的发射光谱不与任何常见的荧光团重叠,从而为流式细胞术中的多色应用提供了独特的颜色。 ReadiUse 预活化的APC-iFluor 800串联蛋白是一种活化的APC蛋白,可以轻松偶联到抗体上,具有比常规APC串联更高的结合产率。

产品说明书

样品分析方案

使用Buccutite MTA制备预激活的抗体

1.在DMSO中以约10 mg / mL的浓度重新配制Buccutite MTA。
注意,请将未使用的Buccutite MTA储存在-20°C下,并且最多可以使用两个冷冻和融化循环。
2.制备浓度大于1 mg / mL的pH = 8.5-9.0缓冲液中的目标抗体(Ab)。
3.以8-10 µg Buccutite MTA / 100 µg Ab的比例将Buccutite MTA添加到Ab溶液中。
4.充分混合并在室温下反应60分钟,在反应过程中旋转。
5.用脱盐柱纯化反应混合物,以除去未反应的Buccutite MTA,并将缓冲液更换为PBS或您选择的缓冲液。
6.收集Buccutite MTA活化的Ab,并以原始起始量的70%产率估算浓度。

 

预激活的APC-iFluor 800串联染料

1.在100 µL ddH2O中将预活化的APC-iFluor 800串联染料重构为10 mg / mL。
注意,重建的预激活APC-iFluor 800串联染料可以在4°C下保存一个月,并避免光照。
2.以130 µg APC-iFluor 800串联染料 / 100 µg MTA激活的Ab的比例直接将APC-iFluor 800串联染料加入MTA激活的目标Ab溶液中。
3.在室温下将混合物离心60分钟。
4.Ab / APC-iFluor 800串联缀合物已经可以使用了。
注意,抗体缀合物应在载体蛋白(例如0.1%牛血清白蛋白)和0.02%-0.05%叠氮化钠存在的情况下以> 0.5 mg / mL的浓度保存。 Ab / APC-iFluor 800串联缀合物溶液可以在4°C下保存长达两个月,并避免光照。
(可选)Ab / APC-iFluor 800串联缀合物可以通过尺寸排阻色谱法(SEC)进一步纯化以获得最佳性能。

荧光素醛是可以与胺,肼或羟胺反应的反应性荧光染料。荧光素是使用最广泛的绿色荧光染料。醛基可与pH 5-9的胺,酰肼或羟胺基反应。与胺反应性琥珀酰亚胺基酯基(NHS)不同,醛可以在酸性pH下与N端胺基反应,这是某些生物缀合反应有时需要的条件。醛与胺基反应形成席夫碱(Schiff base)。用氢化物进一步还原将形成稳定的C-N键。醛与其他基团之间的反应可实现荧光素染料的位点特异性缀合和标记,使其靶向分子上的所需位置。共轭荧光素染料可以很容易地通过普通的荧光仪器在FITC通道下检测到。
